This variant lies in the CFTR gene (transcript NM_000492.4) at coding-DNA position 3319, where T is replaced by C; at the protein level this means replaces phenylalanine at residue 1107 with leucine — a missense variant. Submitter rationale: Variant summary: CFTR c.3319T>C (p.Phe1107Leu) results in a non-conservative amino acid change located in the ABC transporter type 1, transmembrane domain (IPR011527) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 250986 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.3319T>C has been reported in the literature in three individuals affected with Cystic Fibrosis, however in two of such cases, additional variants in CFTR have been reported and the phase relationship with the current variant was unclear (Prach_2013, Raraigh_2021, Salinas_2016). These data do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
