Likely pathogenic for Angelman syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_130839.5(UBE3A):c.449T>C (p.Ile150Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 130 of the UBE3A protein (p.Ile130Thr). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with Angelman syndrome (PMID: 15054837). In at least one individual the variant was observed to be de novo. This variant is also known as 975T>C. ClinVar contains an entry for this variant (Variation ID: 7969). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt UBE3A protein function with a positive predictive value of 80%. Experimental studies have shown that this missense change affects UBE3A function (PMID: 26255772, 33607653, 34815418). In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
