This variant lies in the BRCA1 gene (transcript NM_007294.4) at 14 bases into the intron immediately before coding-DNA position 81, where C is replaced by A. Submitter rationale: The BRCA1 c.81-14C>A variant was not identified in the literature, nor was it identified in dbSNP, the 1000 Genomes Project, the NHLBI Exome Sequencing Project, the Exome Aggregation Consortium, GeneInsight COGR, ClinVar, Clinvitae, COSMIC, MutDB, BRCA Share, BIC, ARUP Laboratories BRCA Mutations Database, the Fanconi Anemia Mutation Database (LOVD). The variant occurs outside of the splicing consensus sequence and 3 of 5 in silico or computational prediction software programs (SpliceSiteFinder, MaxEntScan, NNSPLICE, GeneSplicer, HumanSpliceFinder) predict a greater than 10% difference in splicing. However, this information is not predictive enough to assume pathogenicity.
