This variant lies in the CYP1B1 gene (transcript NM_000104.4) at coding-DNA position 694, where G is replaced by C; at the protein level this means replaces glycine at residue 232 with arginine — a missense variant. Submitter rationale: Variant summary: CYP1B1 c.694G>C (p.Gly232Arg) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 244486 control chromosomes (gnomAD). c.694G>C has been reported in the literature in individuals affected with Primary Congenital Glaucoma or Primary open-angle glaucoma (Colomb_2003, Melki_2004). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
