Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006941.4(SOX10):c.752C>A (p.Ser251Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Ser251*) in the SOX10 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 216 amino acid(s) of the SOX10 protein. This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with Waardenburg syndrome (PMID: 10762540). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 7399). This variant disrupts a region of the SOX10 protein in which other variant(s) (p.Gln399*) have been determined to be pathogenic (PMID: 28000701, 31427586; internal data). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
