NM_000548.5(TSC2):c.2788A>G (p.Lys930Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K930E variant (also known as c.2788A>G), located in coding exon 24 of the TSC2 gene, results from an A to G substitution at nucleotide position 2788. The lysine at codon 930 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
