This variant lies in the RYR1 gene (transcript NM_000540.3) at coding-DNA position 7446, where T is replaced by C; at the protein level this means the protein sequence is unchanged (aspartic acid at residue 2482 retained) — a synonymous variant. Submitter rationale: Variant summary: RYR1 c.7446T>C (p.Asp2482Asp) alters a non-conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. 3/4 computational tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 1.6e-05 in 251222 control chromosomes (gnomAD).
