NM_001999.4(FBN2):c.6293-8T>A was classified as Benign by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: FBN2 c.6293-8T>A alters a non-conserved nucleotide located at a position not widely known to affect splicing. Several computational tools predict a significant impact on normal splicing: Two predict the variant weakens a 3' acceptor site. One predicts the variant abolishes a 3' acceptor site. One predicts the variant has no significant impact on splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 6.7e-05 in 1613178 control chromosomes, predominantly at a frequency of 9.1e-05 within the Non-Finnish European subpopulation in the gnomAD database. The observed variant frequency within Non-Finnish European control individuals in the gnomAD database is approximately 72.8 fold of the estimated maximal expected allele frequency for a pathogenic variant in FBN2 causing Aortopathy phenotype (1.3e-06). To our knowledge, no occurrence of c.6293-8T>A in individuals affected with Aortopathy and no experimental evidence demonstrating its impact on protein function have been reported.
