Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_152296.5(ATP1A3):c.724+8G>A, citing LabCorp Variant Classification Summary - May 2015: Variant summary: ATP1A3 c.724+8G>A alters a nucleotide located at a position not widely known to affect splicing. Several computational tools predict a significant impact on normal splicing: Two predict the variant creates/strengthens a cryptic 5' donor site. One predicts the variant has no significant impact on splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 3.6e-05 in 251442 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.724+8G>A in individuals affected with ATP1A3-Related Disorders and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 696637). Based on the evidence outlined above, the variant was classified as uncertain significance.
