This amino acid residue is located within the BRICHOS domain, which has a role in folding and trafficking the SP-C protein. Two bioinformatic tools queried predict that this substitution would be probably damaging; however, these tools are optimized to predict loss of function variants. As SFTPC variants are thought to act in a dominant negative manner, it is unclear how reliable bioinformatic algorithms would be making predictions for this gene. In addition, the leucine at this position is conserved across most, but not all, mammals, and not across lower species (although some residues in this region are highly conserved). The clinical significance ofthis variant is uncertain.
