Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_017636.4(TRPM4):c.2264G>A (p.Arg755His), citing Ambry Variant Classification Scheme 2023: The p.R755H variant (also known as c.2264G>A), located in coding exon 17 of the TRPM4 gene, results from a G to A substitution at nucleotide position 2264. The arginine at codon 755 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
