Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006361.6(HOXB13):c.634G>C (p.Ala212Pro), citing Ambry Variant Classification Scheme 2023: The p.A212P variant (also known as c.634G>C), located in coding exon 2 of the HOXB13 gene, results from a G to C substitution at nucleotide position 634. The alanine at codon 212 is replaced by proline, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
