Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006361.6(HOXB13):c.578C>G (p.Pro193Arg), citing Ambry Variant Classification Scheme 2023: The p.P193R variant (also known as c.578C>G), located in coding exon 1 of the HOXB13 gene, results from a C to G substitution at nucleotide position 578. The proline at codon 193 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
