Pathogenic variants in this gene have been associated with autosomal dominant developmental and epileptic encephalopathy 79. This variant likely occurred de novo in the current proband and an individual reported in the published literature; however, the possibility of parental germline mosaicism cannot be excluded (PMID: 31056671 ) (PS2_Moderate). An alternate amino acid change at this position (p.Val294Leu) has been previously reported in similarly affected individuals, which suggests that this residue is biologically important (PMID: 29961870) (PM5) and multiple computational algorithms predict a deleterious effect for this variant (REVEL score: 0.812) (PP3). This variant is absent from control populations (https://gnomad.broadinstitute.org/) (PM2).
