Uncertain significance for Granulomatous disease, chronic, autosomal recessive, cytochrome b-positive, type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000433.4(NCF2):c.1105G>A (p.Gly369Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine with arginine at codon 369 of the NCF2 protein (p.Gly369Arg). The glycine residue is highly conserved and there is a moderate physicochemical difference between glycine and arginine. This variant is present in population databases (rs137854513, ExAC 0.02%). This variant has not been reported in the literature in individuals affected with NCF2-related conditions. ClinVar contains an entry for this variant (Variation ID: 68484). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The arginine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
