For these reasons, this variant has been classified as Pathogenic. Experimental studies have shown that this variant affects the structure and function of the alpha-crystallin protein (PMID: 22045060, 22347476, 22140512, 23379525, 25018622, 25694240, 28179137). This variant has been reported to segregate in two families affected with bilateral punctate lenticular opacities and congenital cataracts associated with microcornea (PMID: 17937925, 18587492). Both individuals who were heterozygous for the variant and individuals who were homozygous for the variant were affected. ClinVar contains an entry for this variant (Variation ID: 68457). This sequence change replaces arginine with cysteine at codon 54 of the CRYAA protein (p.Arg54Cys). The arginine residue is highly conserved and there is a large physicochemical difference between arginine and cysteine.
