Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_004360.5(CDH1):c.832+6T>C, citing Sema4 Curation Guidelines: The CDH1 c.832+6T>C variant has not been reported in the literature to our knowledge. It was not observed in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has been reported in ClinVar (Variation ID 681578). The algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant is not likely to affect RNA splicing, though these predictions have not been confirmed by transcriptional studies. The overall evidence is inconsistent with ACMG/AMP requirements for a classification of benign or pathogenic. In summary, the clinical significance of this variant is currently uncertain.
