This variant is present in population databases (rs154774638, gnomAD 0.007%). This missense change has been observed in individual(s) with neuronal ceroid lipofuscinosis (PMID: 21549341). ClinVar contains an entry for this variant (Variation ID: 68096). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt CLN6 protein function with a negative predictive value of 80%. Experimental studies are conflicting or provide insufficient evidence to determine the effect of this variant on CLN6 function (PMID: 32171521). This variant disrupts the p.Arg149 amino acid residue in CLN6.
