Uncertain significance for Long QT syndrome 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_172201.2(KCNE2):c.193G>A (p.Val65Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 65 of the KCNE2 protein (p.Val65Met). This variant is present in population databases (rs199473364, gnomAD 0.006%). This missense change has been observed in individual(s) with clinical features of long QT syndrome (PMID: 12185453, 24606995, 28794082). ClinVar contains an entry for this variant (Variation ID: 67611). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change affects KCNE2 function (PMID: 12185453). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
