This variant lies in the KCNH2 gene (transcript NM_000238.4) at coding-DNA position 1705, where T is replaced by C; at the protein level this means replaces tyrosine at residue 569 with histidine — a missense variant. Submitter rationale: This sequence change replaces tyrosine, which is neutral and polar, with histidine, which is basic and polar, at codon 569 of the KCNH2 protein (p.Tyr569His). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of long QT syndrome (PMID: 10862094; internal data). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 67245). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change affects KCNH2 function (PMID: 25417810).
