This variant lies in the KCNH2 gene (transcript NM_000238.4) at coding-DNA position 1478, where A is replaced by C; at the protein level this means replaces tyrosine at residue 493 with serine — a missense variant. Submitter rationale: This variant is not present in population databases (ExAC no frequency). This sequence change replaces tyrosine with serine at codon 493 of the KCNH2 protein (p.Tyr493Ser). The tyrosine residue is highly conserved and there is a large physicochemical difference between tyrosine and serine.
