Uncertain significance for Cardiac arrhythmia — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000218.3(KCNQ1):c.878G>A (p.Arg293His), citing ACMG Guidelines, 2015. This variant lies in the KCNQ1 gene (transcript NM_000218.3) at coding-DNA position 878, where G is replaced by A; at the protein level this means replaces arginine at residue 293 with histidine — a missense variant. Submitter rationale: This missense variant replaces arginine with histidine at codon 293 of the KCNQ1 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an unaffected control individual in a long QT syndrome case-control study (PMID:19841300). This variant has been identified in 4/250276 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
