Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_013352.4(DSE):c.-53-4G>A, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the DSE gene (transcript NM_013352.4) at 4 bases into the intron immediately before 53 bases upstream of the translation start (5' untranslated region), where G is replaced by A. Submitter rationale: Variant summary: DSE c.-53-4G>A is located in the untranslated mRNA region upstream of the initiation codon. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 3.7e-05 in 246116 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.-53-4G>A in individuals affected with Ehlers-Danlos syndrome, musculocontractural type 2 and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 669056). Based on the evidence outlined above, the variant was classified as uncertain significance.
