Pathogenic for Charcot-Marie-Tooth disease type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_170707.4(LMNA):c.357-1G>T, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the LMNA gene (transcript NM_170707.4) at the canonical splice acceptor site of the intron immediately before coding-DNA position 357, where G is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects an acceptor splice site in intron 1 of the LMNA gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or disrupted protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individuals with dilated cardiomyopathy (PMID: 18585512, 27182706). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 66893). Studies have shown that disruption of this splice site results in skipping of exon 2 and introduces a premature termination codon (PMID: 18585512). The resulting mRNA is expected to undergo nonsense-mediated decay. For these reasons, this variant has been classified as Pathogenic.
