This variant lies in the LMNA gene (transcript NM_170707.4) at coding-DNA position 11, where C is replaced by G; at the protein level this means replaces proline at residue 4 with arginine — a missense variant. Submitter rationale: This sequence change replaces proline with arginine at codon 4 of the LMNA protein (p.Pro4Arg). The proline residue is highly conserved and there is a moderate physicochemical difference between proline and arginine. While this variant is not present in population databases, the frequency information is unreliable, as metrics indicate poor data quality at this position in the ExAC database. This variant has been observed to be de novo in individuals affected with atypical progeroid syndrome and to segregate with disease in affected families (PMID: 19875478, 24861648). ClinVar contains an entry for this variant (Variation ID: 66795).
