NM_001378454.1(ALMS1):c.7217C>T (p.Pro2406Leu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P2407L variant (also known as c.7220C>T), located in coding exon 8 of the ALMS1 gene, results from a C to T substitution at nucleotide position 7220. The proline at codon 2407 is replaced by leucine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
