Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000245.4(MET):c.3865A>T (p.Thr1289Ser), citing Ambry Variant Classification Scheme 2023: The p.T1307S variant (also known as c.3919A>T), located in coding exon 19 of the MET gene, results from an A to T substitution at nucleotide position 3919. The threonine at codon 1307 is replaced by serine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
