Pathogenic for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_014845.6(FIG4):c.1928T>A (p.Leu643Ter), citing Ambry Variant Classification Scheme 2023: The p.L643* pathogenic mutation (also known as c.1928T>A), located in coding exon 17 of the FIG4 gene, results from a T to A substitution at nucleotide position 1928. This changes the amino acid from a leucine to a stop codon within coding exon 17. This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
