This variant lies in the MPL gene (transcript NM_005373.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 981, where G is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: MPL c.981-1G>C is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Several computational tools predict a significant impact on normal splicing: Three predict the variant abolishes a 3 acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 1.2e-05 in 251340 control chromosomes. c.981-1G>C has been not been reported in the literature in individuals affected with Congenital Amegakaryocytic Thrombocytopenia.
