Uncertain Significance for Familial isolated arrhythmogenic right ventricular dysplasia — the classification assigned by All of Us Research Program, National Institutes of Health to NM_024422.6(DSC2):c.874C>T (p.Pro292Ser), citing ACMG Guidelines, 2015. This variant lies in the DSC2 gene (transcript NM_024422.6) at coding-DNA position 874, where C is replaced by T; at the protein level this means replaces proline at residue 292 with serine — a missense variant. Submitter rationale: Variant of Uncertain Significance due to insufficient evidence: This missense variant is located in the extracellular cadherin domain 2 of the DSC2 protein. Computational prediction tools and conservation analyses are inconclusive regarding the impact of this variant on the protein function. Computational splicing tools suggest that this variant may not impact RNA splicing. To our knowledge, functional assays have not been performed for this variant nor has this variant been reported in individuals affected with cardiovascular disorders in the literature. This variant has been identified in 5/277168 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Available evidence is insufficient to determine the pathogenicity of this variant conclusively.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
