While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 19 amino acid(s) of the CRYBA1 protein and extend the protein by 2 additional amino acid residues. This variant is present in population databases (no rsID available, gnomAD 0.0009%). This frameshift has been observed in individuals with autosomal dominant congenital cataract (PMID: 24926697, 37458831; internal data). It has also been observed to segregate with disease in related individuals. For these reasons, this variant has been classified as Pathogenic.
