Uncertain significance for Progressive myoclonic epilepsy type 3 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_153033.5(KCTD7):c.636T>A (p.Phe212Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine with leucine at codon 212 of the KCTD7 protein (p.Phe212Leu). The phenylalanine residue is highly conserved and there is a small physicochemical difference between phenylalanine and leucine. This variant is present in population databases (rs752360120, ExAC 0.001%). This variant has not been reported in the literature in individuals affected with KCTD7-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
