Uncertain significance for Neuropathy, hereditary sensory and autonomic, type 1C — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004863.4(SPTLC2):c.620G>A (p.Arg207Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 207 of the SPTLC2 protein (p.Arg207Gln). This variant is present in population databases (rs751801159, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with SPTLC2-related conditions. ClinVar contains an entry for this variant (Variation ID: 665330). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt SPTLC2 protein function with a positive predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
