Uncertain significance for Autosomal recessive limb-girdle muscular dystrophy type 2Q; Epidermolysis bullosa simplex, Ogna type; Epidermolysis bullosa simplex with nail dystrophy; Epidermolysis bullosa simplex 5C, with pyloric atresia; Epidermolysis bullosa simplex 5B, with muscular dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_201384.3(PLEC):c.9368G>A (p.Gly3123Asp), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the PLEC gene (transcript NM_201384.3) at coding-DNA position 9368, where G is replaced by A; at the protein level this means replaces glycine at residue 3123 with aspartic acid — a missense variant. Submitter rationale: This variant is not present in population databases (ExAC no frequency). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with PLEC-related disease. This sequence change replaces glycine with aspartic acid at codon 3150 of the PLEC protein (p.Gly3150Asp). The glycine residue is highly conserved and there is a moderate physicochemical difference between glycine and aspartic acid.
