This variant lies in the RPS24 gene (transcript NM_033022.4) at coding-DNA position 91, where G is replaced by A; at the protein level this means replaces glycine at residue 31 with arginine — a missense variant. Submitter rationale: The p.G31R variant (also known as c.91G>A), located in coding exon 3 of the RPS24 gene, results from a G to A substitution at nucleotide position 91. The glycine at codon 31 is replaced by arginine, an amino acid with dissimilar properties. This variant was reported in an individual with a congenital heart defect and elevated ADA, as well as in the healthy father (Vlachos A et al. Circ Genom Precis Med, 2018 05;11:e002044). Northern blot analysis performed on total RNA isolated from the patient's primary peripheral blood mononuclear cells showed increased 30S pre-rRNA and diminished 18SE compared to an immortalized cell line, consistent with a pre-rRNA processing defect (Vlachos A et al. Circ Genom Precis Med, 2018 05;11:e002044). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on available evidence to date, the clinical significance of this alteration remains unclear.
