NM_004304.5(ALK):c.2999A>G (p.Glu1000Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1000G variant (also known as c.2999A>G), located in coding exon 18 of the ALK gene, results from an A to G substitution at nucleotide position 2999. The glutamic acid at codon 1000 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
