This variant lies in the CNTN2 gene (transcript NM_005076.5) at coding-DNA position 2514, where G is replaced by C; at the protein level this means replaces glutamine at residue 838 with histidine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C15"). This variant has not been reported in the literature in individuals with CNTN2-related disease. This variant is present in population databases (rs144293383, ExAC 0.009%).
