Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.8254G>A (p.Asp2752Asn), citing Ambry Variant Classification Scheme 2023: The p.D2752N variant (also known as c.8254G>A), located in coding exon 64 of the FBN2 gene, results from a G to A substitution at nucleotide position 8254. The aspartic acid at codon 2752 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
