Uncertain significance for Hereditary nonpolyposis colorectal neoplasms — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000179.3(MSH6):c.3033T>G (p.Ile1011Met), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the MSH6 gene (transcript NM_000179.3) at coding-DNA position 3033, where T is replaced by G; at the protein level this means replaces isoleucine at residue 1011 with methionine — a missense variant. Submitter rationale: This sequence change replaces isoleucine with methionine at codon 1011 of the MSH6 protein (p.Ile1011Met). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and methionine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with MSH6-related disease. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
