Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_000368.5(TSC1):c.2432G>A (p.Arg811Gln), citing Sema4 Curation Guidelines. This variant lies in the TSC1 gene (transcript NM_000368.5) at coding-DNA position 2432, where G is replaced by A; at the protein level this means replaces arginine at residue 811 with glutamine — a missense variant. Submitter rationale: The TSC1 c.2432G>A (p.R811Q) variant has not been reported in individuals with TSC1-related disease. It was observed in 1/113676 chromosomes in the Non-Finnish European subpopulation according to the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has been reported in ClinVar (Variation ID: 664903). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
