NM_017636.4(TRPM4):c.3434C>G (p.Ser1145Cys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1145C variant (also known as c.3434C>G), located in coding exon 22 of the TRPM4 gene, results from a C to G substitution at nucleotide position 3434. The serine at codon 1145 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
