This variant lies in the MSH2 gene (transcript NM_000251.3) at coding-DNA position 2401, where C is replaced by T; at the protein level this means replaces histidine at residue 801 with tyrosine — a missense variant. Submitter rationale: The MSH2 p.His801Tyr variant was not identified in the literature nor was it identified in the dbSNP, ClinVar, Genesight-COGR, Cosmic, UMD-LSDB, Insight Colon Cancer Gene Variant Database, Zhejiang Colon Cancer Database, Mismatch Repair Genes Variant Database, Insight Hereditary Tumors Database, the 1000 Genomes Project, the NHLBI GO Exome Sequencing Project or the Exome Aggregation Consortium (August 8th 2016) control databases. The p.His801 residue is conserved across mammals and other organisms, and four out of five computational analyses (PolyPhen-2, SIFT, AlignGVGD, BLOSUM, MutationTaster) suggest that the Tyr variant may impact the protein; however, this information is not predictive enough to assume pathogenicity.
