This variant lies in the SCN1A gene (transcript NM_001165963.4) at coding-DNA position 958, where G is replaced by T; at the protein level this means replaces aspartic acid at residue 320 with tyrosine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This sequence change replaces aspartic acid with tyrosine at codon 320 of the SCN1A protein (p.Asp320Tyr).
