NM_015046.7(SETX):c.4459T>A (p.Ser1487Thr) was classified as Uncertain significance for Amyotrophic lateral sclerosis type 4; Spinocerebellar ataxia, autosomal recessive, with axonal neuropathy 2 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with SETX-related disease. This variant is present in population databases (rs772466321, ExAC 0.001%). This sequence change replaces serine with threonine at codon 1487 of the SETX protein (p.Ser1487Thr). The serine residue is weakly conserved and there is a small physicochemical difference between serine and threonine.
