Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_002439.5(MSH3):c.1340+6T>C, citing Sema4 Curation Guidelines: The MSH3 c.1340+6T>C variant has not been reported in the literature to our knowledge. This variant was observed in 2/34556 chromosomes in the Latino population according to the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID: 664657). In silico tools suggest the variant may not disrupt normal splicing, though these predictions have not been confirmed by functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
