Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_002230.4(JUP):c.148G>A (p.Gly50Arg), citing Ambry Variant Classification Scheme 2023: The p.G50R variant (also known as c.148G>A), located in coding exon 1 of the JUP gene, results from a G to A substitution at nucleotide position 148. The glycine at codon 50 is replaced by arginine, an amino acid with dissimilar properties. This alteration was reported in an arrhythmogenic cardiomyopathy cohort (Rasmussen TB et al. Circ Cardiovasc Genet, 2014 Jun;7:230-40). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
