Uncertain significance for Desmin-related myofibrillar myopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001927.4(DES):c.322G>A (p.Glu108Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 108 of the DES protein (p.Glu108Lys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with dilated cardiomyopathy (PMID: 17325244, 38727660). ClinVar contains an entry for this variant (Variation ID: 66408). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant.
