NM_000465.4(BARD1):c.143T>A (p.Leu48Gln) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L48Q variant (also known as c.143T>A), located in coding exon 1 of the BARD1 gene, results from a T to A substitution at nucleotide position 143. The leucine at codon 48 is replaced by glutamine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
