This variant lies in the PRKAG2 gene (transcript NM_016203.4) at coding-DNA position 679, where A is replaced by G; at the protein level this means replaces lysine at residue 227 with glutamic acid — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This sequence change replaces lysine with glutamic acid at codon 227 of the PRKAG2 protein (p.Lys227Glu).
