Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000249.4(MLH1):c.1849A>G (p.Lys617Glu), citing Ambry Variant Classification Scheme 2023: The p.K617E variant (also known as c.1849A>G), located in coding exon 16 of the MLH1 gene, results from an A to G substitution at nucleotide position 1849. The lysine at codon 617 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
